Zinc · Regulation (EU) No 432/2012

The same ion in the Leydig cell, in the red blood cell, and in the cell nucleus

Anhydritsaum takes the statements authorized for zinc apart one by one and describes the process behind each. Zinc contributes to normal acid-base metabolism — Pursuant to Regulation (EU) No 432/2012.

An element needed in very different places

The EU Commission has authorized several health claims for zinc. Each goes back to a separate opinion, and each describes a different process in the body. We set four of them side by side here.

Zn

The hormone arises in the interstitial tissue of the testis

Between the seminiferous tubules lie the Leydig cells. They convert cholesterol into testosterone through a series of enzymatic steps. Several stations of this chain, and also the overarching hormone release of the pituitary gland, cannot manage without bound zinc ions.

Zn

Fatty acids are remodeled, not merely burned

The organism shortens, lengthens, and desaturates fatty acids depending on what it intends them for: as a membrane building block, as an energy carrier, or as a starting material for messenger substances. At several of these remodeling steps sit enzymes with zinc in the active center.

Zn

The pH of the blood stays within narrow limits

In a healthy person, blood moves between pH 7.35 and 7.45. For this narrow corridor to be maintained, carbon dioxide as it arises must continually be brought into a transportable form. The enzyme that accelerates this conversion many times over carries a zinc ion in its pocket.

Zn

Before every cell division the genome is duplicated

Skin, intestinal lining, and bone marrow renew themselves without interruption, and every new cell needs a complete set of genetic information. The enzymes that string building blocks together and correct reading errors in the process belong to the zinc-dependent proteins of the cell nucleus.

Zinc acts through the shape of proteins

An adult body carries between two and three grams of zinc. That is little measured against calcium, and it is moreover distributed across very many locations: musculature and skeleton hold the largest share, the rest sits in the liver, skin, prostate, retina, and in practically every cell. There is no organ that collects zinc for lean times.

The reason for this broad distribution lies in the way the ion works. It does not accelerate a reaction by reacting itself. It settles into a pocket of the protein molecule, holds several amino acid residues there at a fixed distance from one another, and thereby gives the molecule the fit without which it cannot perform its task. Estimates speak of several hundred enzymes of human metabolism built in this way.

An ion that exchanges no electrons

Iron and copper switch back and forth between two charge states in metabolism; precisely this makes them useful for electron transfers and at the same time demanding to handle. Zinc is present in the body practically always doubly positively charged and remains so. This evenness is the reason it can be deployed at sites where a reactive metal would cause damage — for instance directly at the genome.

Because the ion is stored nowhere, the ongoing requirement depends on the daily diet. The organism adjusts to this: when intake is scarce, the share the intestinal wall lets through rises, and excretion via the digestive tract declines. This readjustment does, however, have a range within which it works.

Zinc finger: a folding pattern that fits into the major groove

In the 1980s, structural biologists described a recurring motif in proteins that bind to DNA: a short amino acid chain that lays itself into a loop around a zinc ion. This loop has exactly the dimensions to reach into the wide turn of the double helix and recognize a particular sequence of letters there.

Such zinc fingers are among the most common components of human transcription factors, that is, of those proteins that determine which gene is read. The enzymes that assemble new DNA strands also work with bound zinc. The EU Commission has recorded this connection in a statement of its own: „Zinc contributes to normal DNA synthesis“ — Pursuant to Regulation (EU) No 432/2012.

How carbon dioxide becomes bicarbonate in the blood

Every cell releases carbon dioxide. Dissolved in the blood, this would become carbonic acid, and the pH would fall. The body solves it differently: in the red blood cells sits an enzyme that converts carbon dioxide and water into bicarbonate and a hydrogen ion in a flash. Without this enzyme the same reaction also proceeds, only orders of magnitude more slowly than circulation demands.

At the center of this enzyme sits a single zinc ion that polarizes a water molecule far enough for it to attack the carbon dioxide. The bicarbonate then travels into the plasma and serves there as a buffer; in the lungs the process runs backward and the gas is breathed out. Over hours, the kidney adjusts what breathing does not balance within minutes.

The pituitary sets the rhythm

The formation of testosterone begins not in the testis but in the head. The hypothalamus releases a control hormone in pulses, the pituitary gland responds with luteinizing hormone, and only this reaches the Leydig cells via the blood. There it sets in motion the conversion of cholesterol, which leads via pregnenolone and several intermediate stages to the finished hormone.

Because zinc-dependent enzymes are involved in this chain and because release in the anterior pituitary likewise depends on the ion, the connection was assessed by the authority and given a formulation of its own: „Zinc contributes to the maintenance of normal testosterone levels in the blood“ — Pursuant to Regulation (EU) No 432/2012. The statement speaks of maintenance, not of increase; that is not a linguistic nicety but the core of the authorization.

Why dietary fat does not stay as it arrives

What lies on the plate as fat is seldom used by the body unchanged. Chains are lengthened, double bonds inserted, other molecules broken down step by step until usable building blocks remain. Whether a fatty acid is built into the cell membrane, processed further into messenger substances, or drawn on for energy production is decided in this remodeling.

Several of the enzyme groups involved carry zinc as a fixed constituent. These include steps of lipid breakdown as well as enzymes involved in the processing of alcohol and other alcohols, which use the same construction type. „Zinc contributes to normal fatty acid metabolism“ — Pursuant to Regulation (EU) No 432/2012.

The difference between a function and a promise

Anyone advertising a food may not ascribe to it an effect that has not been assessed and included in the Union list. This list is the annex to Regulation (EU) No 432/2012. It contains a limited number of statements for each nutrient, together with the conditions under which a product may carry them.

Striking in all the statements is a single word: normal. It marks the scope. What is described is the contribution to a process that runs anyway — not its increase beyond the ordinary measure, and certainly not an effect on an illness. Claims of the second and third kind are not permissible in advertising for foods, regardless of whether they are accurate.

A second condition concerns quantity. A product may carry one of these statements only if it contains an appreciable amount of the nutrient; the yardstick is a share of the reference value, governed by a separate regulation. A food with traces of zinc therefore falls outside, even though the physiology remains the same.

This is why you will find on these pages no symptom pictures, no testimonials, and no recommendation to take anything. You will find the authorized wording, the processes to which it refers, and figures that can be looked up. Everything beyond that belongs in a conversation with a physician.
